Ligninolytic Enzyme Production and Remazol Brilliant Blue R (rbbr) Decolorization by a Newly Isolated White Rot Fungus: Basidiomycota Spp. L-168

چکیده
Recently, extensive research on basidiomycetous fungi has been conducted to isolate new organisms with enormous secretion of ligninolytic enzymes with potential industrial applications. The present study reports the screening of 13 phenol tolerant isolates from 51 rotted wood samples collected from Jalgaon district, Maharashtra (India). The isolates were tested for ligninolytic enzyme production by a two step screening based on phenol tolerance and gallic acid oxidation. Also ability of isolates for Remazol brilliant blue R decolorization was studied. Interestingly, Basidiomycete sp. L-168 showed 98.53% decolorization in 5 days of incubation. The isolate was characterized with respect to its set of extracellular phenol oxidases. Laccase was the only ligninolytic enzyme detected. Neither lignin peroxidase nor manganese peroxidase was detected. Maximum of 507.92 U l of laccase was produced on 7 day at 30C. This organism appears to be an ideal model for studies of group of fungi that lack LiP and MnP and operate with laccase as the major phenol oxidase. This is first report on isolation of ligninolytic fungi from Jalgaon district.

Ligninolytic Enzymes Production and Remazol Brilliant Blue R Decolorization by Tropical Brazilian Basidiomycetes Fungi
Remazol Brilliant Blue R (RBBR) dye was used as substrate to evaluate ligninolytic activity in 125 basidiomycetous fungi isolated from tropical ecosystems. The extracellular RBBR decolorizing activity produced when selected fungi were grown in solid media and in soil contaminated with organochlorines was also evaluated. A total of 106 fungi decolorized the RBBR during the growth in malt extract...
متن کاملUse of sugarcane molasses by Pycnoporus sanguineus for the production of laccase for dye decolorization.
Pycnoporus sanguineus is a white-rot basidiomycete that produces laccase as the only oxidoreductase; enzyme synthesis depends on cultivation variables, and fungal species and strain. Laccases have wide substrate specificity, oxidize a broad range of compounds, and show potential for use in dye decolorization. We evaluated laccase production in a recently isolated strain of P. sanguineus cultiva...

Response surface methodology (central composite design of experiments) was employed to simultaneously optimize enzyme production and productivities of two ligninolytic enzymes produced by Ceriporiopsis subvermispora. Concentrations of glucose, ammonium tartrate and Polysorbate 80 were varied to establish the optimal composition of liquid media (OLM), where the highest experimentally obtained ac...

(Cultural and ligninolytic activity studies of some polypores (Basidiomycota) from brazilian Atlantic Forest, São Paulo State, Brazil). During a fungi survey in the brazilian Atlantic Forest in São Paulo, São Paulo State, Brazil, polypores growing on decaying wood were collected and fragments of the basidiomata were used to obtain isolates in pure culture. A total of 37 isolates were obtained a...
متن کاملDecolourisation of Remazol Brilliant Blue R via a novel Bjerkandera sp. strain.
A novel strain of Bjerkandera sp. (B33/3), with particularly high decolourisation activities upon Poly R-478 and Remazol Brilliant Blue R (RBBR) dyes, was isolated. The role of the ligninolytic extracellular enzymes produced by this strain on decolourisation of RBBR was studied in some depth. The basis of decolourisation is an enzyme-mediated process, in which the main enzyme responsible is a r...
